My connection was working fine through wi-fi but it was not working for USB. So, I installed HTC Sync and now it is working for USB but it is not working for Wifi now.

Did you start the client on your Desire before you tried to connect?
With USB connection the client starts itselfs on the phone, but not with WiFi by default, if you did not set that option inside the client settings.
